First up, and just to make sure we understand, you're able to see what you're playing on Spotify from Discord on your mobile and the web, but not from the Discord desktop app?
If that's the case, head to User Settings > Connections and then you can toggle Display on Profile and Display Spotify as your status. You can also try reinstalling the app to check if that makes a difference.
Lastly, could you send a pic of how the status shows on your computer? We'll take a look.
I have turned on the playback status in your profile as you mentioned. Currently, except for the Windows version of Spotify, other versions (web, Android) can connect to the Discord playback status. Windows cannot detect the device, while other versions can detect it.

Just to make sure we cover all bases, do you notice any difference after a clean reinstall of the desktop app? This will clear any stored & damaged cache that might interfere with the app, as well as ensure that you're running the latest app version.
If that doesn't do the trick, could you let us know if you previously were able to see the playback status on the computer app? If so, did you notice that it disappeared at any specific point? For example, after an update to the Spotify app or to Discord.

The exact same thing happens to me and I followed the steps, I reinstalled Discord and Spotify and still nothing, it's as if the Spotify app is offline, it doesn't show up in Discord and in the mobile app I can't connect to the PC
Since I first installed the computer version of Spotify, I have been unable to sync the playback status and the device being played. However, my playlists and other things can be kept in sync, but the playback status cannot be synced

Thanks for keeping in touch.
For starters, we recommend rebooting your router/modem. You can also try with a different internet connection and check if that makes a difference. A mobile hotspot is usually a convenient way to test this.
It's also a good idea to go in the Desktop app's Settings and double check if proxy settings are set to default (auto-detect). Additionally, try adding Spotify as an exclusion to the Antivirus, Firewall and/or Windows Security on your computer. You'll find the steps for the last one on the Microsoft Support page.
Lastly, if you're using a VPN, we'd recommend turning it off to check if it could be related to some network restrictions.
